Involvement of Endothelin-1 in Adrenal Catecholamine Regulation

چکیده
Aims: The purpose of the current study was to evaluate the significance of the endothelin system in catecholamine release and synthesis in the adrenal gland. Main Methods: Primary cultures of isolated bovine adrenal medullary cells were prepared. Gene expression, intracellular calcium changes, epinephrine release, and other factors were investigated. Key findings: Expression of endothelin receptors in the rat adrenal gland was confirmed. Intravenous infusion of endothelin-1 (1.0 nmol) increased blood pressure (systolic and diastolic). Endothelin-1 stimulated intracellular calcium changes, resulting in increased nuclear factor of T cell (NFAT) activity and epinephrine release from cultured adrenal medullary cells. Furthermore, endothelin-1 increased catecholamine synthesis and caused hypertrophic changes in the cell size. Significance: Our findings indicate involvement of the endothelin system in the sympathetic regulation of the adrenal medulla. Hirosaki Med.J. 65:218―226,2014

Role of ET(B) receptors and nitric oxide in adrenal catecholamine secretion in anesthetized dogs.
We examined the effects of sarafotoxin 6c (S6c), an endothelin-B (ET(B)) receptor agonist, on adrenal catecholamine secretion in response to cholinergic stimuli in pentobarbital sodium-anesthetized dogs. Drugs were administered intra-arterially into the adrenal gland through the phrenicoabdominal artery. Infusion of S6c attenuated increases in adrenal catecholamine output induced by splanchnic ...
متن کاملEndothelin enhances and inhibits adrenal catecholamine release in deoxycorticosterone acetate-salt hypertensive rats.
Endothelin (ET) and the sympathoadrenal system contribute to the development and maintenance of deoxycorticosterone acetate (DOCA)-salt hypertension. ET can act directly on the adrenal medulla to enhance the release of catecholamines. In addition, the level of ET peptide is increased in the adrenal glands of DOCA-salt hypertensive rats. Therefore, we tested the hypothesis that ET enhances adren...
متن کاملRole of endothelin-1 in stress response in the central nervous system.
Endothelin (ET)-1 is a 21-amino acid peptide that induces a variety of biological activities, including vasoconstriction and cell proliferation, and its likely involvement in cardiovascular and other diseases has recently led to broad clinical trials of ET receptor antagonists. ET-1 is widely distributed in the central nervous system (CNS), where it is thought to regulate hormone and neurotrans...
متن کاملInterplay of Phosphorylated Apoptosis Repressor with CARD, Casein Kinase-2 and Reactive Oxygen Species in Regulating Endothelin-1–Induced Cardiomyocyte Hypertrophy
Objective(s): The role of the Apoptosis repressor with caspase recruitment domain (ARC) in apoptosis and in certain hypertrophic responses has been previously investigated, but its regulation of Endothelin-1 induced cardiac hypertrophy remains unknown. The present study discusses the inhibitory role of ARC against endothelin–induced hypertrophy. Results:In present study Endothelin treated car...
متن کاملRegulation of plasma endothelin by salt in salt-sensitive hypertension.
BACKGROUND Salt dependency of blood pressure (BP) characterizes most models of experimental hypertension in which endothelins play a significant vasoconstrictor role. Despite this, there are no data on the regulation of plasma endothelin by salt balance in human hypertension. METHODS AND RESULTS Plasma endothelin was measured in 47 patients with essential hypertension. Endothelin, catecholami...
